This project’s goal is to explore the use of a novel method of erythrocyte engineering, short-term incubation with gold nanoparticles, to label erythrocytes for the purpose of developing a novel method for imaging blood flow dynamics. Blood flow is known to be intimately related to the survival of animal cells. It has also been shown that blood flow can be manipulated in the treatment of cancer, infection, or other diseases. Our goal is to improve the method used to label erythrocytes for the purposes of developing a powerful tool to image blood flow dynamics. In this project, the aim is to label the blood cells with quantum dots (Q-dots) or gold nanoparticles to create a novel method to image erythrocyte blood flow dynamics and correlate blood flow to cell function. This technology could be applied to a wide variety of diseases. One disease in which it might be particularly useful is cancer. Targeted delivery of drugs to solid tumors depends on the ability of drugs and other therapeutic agents to enter the tumor cells and not be carried away by the blood flow. Because the endothelial cells lining the tumor vasculature are defective, drugs that enter the blood and then the general circulation may be carried away by the circulating blood flow into the tumor.
